Use of 18F-2-Fluorodeoxyglucose to Label Antibody Fragments for Immuno-Positron Emission Tomography of Pancreatic Cancer

چکیده
We generated F-labeled antibody fragments for positron emission tomography (PET) imaging using a sortase-mediated reaction to install a trans-cyclooctenefunctionalized short peptide onto proteins of interest, followed by reaction with a tetrazine-labeled-F-2-deoxyfluoroglucose (FDG). The method is rapid, robust, and site-specific (radiochemical yields > 25%, not decay corrected). The availability of F-2-deoxyfluoroglucose avoids the need for more complicated chemistries used to generate carbon− fluorine bonds. We demonstrate the utility of the method by detecting heterotopic pancreatic tumors in mice by PET, using anti-Class II MHC single domain antibodies. We correlate macroscopic PET images with microscopic two-photon visualization of the tumor. Our approach provides easy access to Flabeled antibodies and their fragments at a level of molecular specificity that complements conventional F-FDG imaging.

Objective(s): To investigate the diagnostic accuracy of positron emission mammography (PEM) and positron emission tomography/computed tomography (PET/CT) for small breast tumors of less than 20 mm in size. Methods: The study was conducted on a total of 100 subjects (i.e., 50 patients with pathologically proven breast cancer and 50 normal cases of medical screening). The total number of tumors wa...
متن کاملPredictive ability of 18F-fluorodeoxyglucose positron emission tomography/computed tomography for pathological complete response and prognosis after neoadjuvant chemotherapy in triple-negative breast cancer patients
Objective The mortality of patients with locally advanced triple-negative breast cancer (TNBC) is high, and pathological complete response (pCR) to neoadjuvant chemotherapy (NAC) is associated with improved prognosis. This retrospective study was designed and powered to investigate the ability of 18F-fluorodeoxyglucose positron emission tomography/computed tomography (FDG-PET/CT) to predict pat...
متن کاملBackground-Based Delineation of Internal Tumor Volumes on Static Positron Emission Tomography in a Phantom Study
Objective(s): Considering the fact that the standardized uptake value (SUV) of a normal lung tissue is expressed as x±SD, x+3×SD could be considered as the threshold value to outline the internal tumor volume (ITV) of a lung neoplasm. Methods: Three hollow models were filled with 55.0 kBq/mL fluorine18- fluorodeoxyglucose (18F-FDG) to represent tumors. The models were fixed to a barrel filled w...
متن کاملBispecific antibody pretargeting of radionuclides for immuno single-photon emission computed tomography and immuno positron emission tomography molecular imaging: an update.
Molecular imaging is intended to localize disease based on distinct molecular/functional characteristics. Much of today's interest in molecular imaging is attributed to the increased acceptance and role of 18F-flurodeoxyglucose (18F-FDG) imaging in a variety of tumors. The clinical acceptance of 18F-FDG has stimulated research for other positron emission tomography (PET) agents with improved sp...
متن کاملEvaluation of Simultaneous Dual-radioisotope SPECT Imaging Using 18F-fluorodeoxyglucose and 99mTc-tetrofosmin
Objective(s): Use of a positron emission tomography (PET)/single-photonemission computed tomography (SPECT) system facilitates the simultaneousacquisition of images with fluorine-18 fluorodeoxyglucose (18F-FDG) andtechnetium (99mTc)-tetrofosmin. However, 18F has a short half-life, and 511keV Compton-scattered photons are detected in the 99mTc energy window. Therefore, in this study, we aimed to ...
